Ketoconazole API
An antifungal drug called ketoconazole is used to treat specific fungus-related illnesses.
Ketoconazole is an imidazole antifungal drug that is effective against many different types of fungus.
Increased membrane fluidity and suppression of fungus development result from its ability to inhibit the production of ergosterol, the fungal counterpart of cholesterol.
In 1981, the Food and Drug Administration (FDA) green-lighted oral ketoconazole for systemic use.
Fungal infections such as histoplasmosis, chromomycosis, and paracoccidioidomycosis, as well as others, can be treated or prevented using ketoconazole. Label It is also used to treat endogenous Cushing’s syndrome in Europe.
Like other azole antifungals, ketoconazole is a fungistatic drug, meaning it stops the growth of fungal cells and prevents their spread.
Ketoconazole inhibits the cytochrome P-450 enzyme 14–sterol demethylase, which is required for the biotransformation of lanosterol into ergosterol.5,11 Reduced levels of ergosterol in the fungal cell membrane lead to ergosterol production inhibition and increased cellular permeability. The formation of the hazardous metabolite 14-methyl-3,6-diol is another consequence of this metabolic restriction. Since the membrane’s components are no longer packed as tightly, it is believed that this leads to an impairment of membrane-bound enzyme systems.
