Closantel Base

An antiparasitic active element known as Closantel is used in veterinary medicine to treat both internal and external parasites that affect livestock, such as sheep nasal bots, cattle grubs, and several kinds of roundworms and liver flukes. Pests in homes and agriculture are not combated by it. It belongs to the salicylanilides chemical class.

Liver flukes, specific gastrointestinal roundworms (= nematodes), and various myiases (= parasitic maggots) are the main veterinary parasites handled.

Closantel and other salicylanilides have a partially understood molecular mechanism of action. They are all uncouplers of oxidative phosphorylation, which disrupts the process by which ATP, the cellular “fuel,” is produced in the mitochondria of the cell. This appears to happen via inhibiting the activity of the enzymes involved in this process, succinate dehydrogenase and fumarate reductase. The parasite’s motility is affected, along with possibly other processes. It appears that closantel also interferes with the parasite membranes’ ion and liquid transportation systems.

Closantel has recently been found to suppress the chitinase enzyme in Onchocerca volvulus, a filarial nematode that causes river blindness in humans. An enzyme called chitinase helps larvae moult. Its inhibition stops them from maturing into adult worms.
